Nanoparticles Stabilized with MPEG-Polylysine Carbamate: Synthesis and Characterization

Biocompatible polymer-coated nanoparticles are at the forefront of research into the ever expanding field of drug delivery. We synthesized a reactive O-imidazolide precursor of methoxy polyethylene glycol (MPEG) and linked it to poly-L-lysine of two separate molecular masses to synthesize biocompatible protected graft copolymers (MPEG-gPLL). MPEG-gPLLs were then used for coating gold nanoparticles obtained in the presence of citrate as a capping/reducing reagent. We also performed the assessment of stabilization using surface plasmon changes and fluorescence spectroscopy and determined the appropriate concentrations of PGC to be between 0.0156 and 0.004 mg/ml and nanoparticles to be 0.99 mg/ml for optimal binding to the surface of the nanoparticles.
