Hyperspectral Fluorescence Imaging for Molecular Biology Applications

Current fluorescence microscopes are unable to discriminate fluorophores emit in the same color channel. Hyperspectral sensors can capture the shape of emission spectra unique to each fluorophore. Our project aimed at using the power of hyperspectral imaging to provide a higher resolution for fluorescence microscopy. Such a system allows researchers to expand the repertoire of probes used in the multiplex analysis. To achieve our project objectives, we designed a light source using LEDs in the 375 to 780 nm range. Additionally, we developed a graphical user interface for users to control LEDs. By using spectral analysis, including linear unmixing and spectral angle mapper, our device combine with a hyperspectral sensor, we were able to identify fluorophores emit in the same color channel.
