Design of a Cell Delivery Vehicle for Volumetric Muscle Loss

The purpose of this project was to design, produce, and test a cell delivery vehicle to aid in the treatment of volumetric muscle loss. Fibrin, collagen I, laminin-111, chitosan, fibronectin, and vitronectin were tested on a two dimensional (2D) scale. Compared to fibrin, there was a statistically higher percent attachment for collagen I and these adhering cells were found to have a higher myogenic index at the 2D scale. Effective collagen I coating of the fibrin microthread bundle was verified through immunocytochemistry. The collagen I coated fibrin microthread preserves the axial alignment of the current system and was found to have higher human satellite cell attachment without causing premature differentiation or cell death, when compared to the uncoated fibrin microthread bundle.
