Histone Occupancy of PRMT8 5’ UTR in Human Dermal Fibroblasts during the Acquisition of iRC Phenotype

PRMT8 is a chromatin modification enzyme which catalyzes arginine methylation reactions. Previous research shows culture conditions alone (2% oxygen and FGF2 supplementation) can activate transcription of PRMT8 in human dermal fibroblasts. We hypothesize that changes in histone occupancy of the PRMT8 5’ UTR are associated with changes in PRMT8 expression. Chromatin Immunoprecipitation was used to assess histone H4ac and histone H3K9me2 occupancy of the PRMT8 5’ UTR that may be linked to activation and/or repression of PRMT8 transcription. We have demonstrated that PRMT8 gene expression may be regulated by differential occupancy of PRMT8 5’UTR region by differently modified histones H3 and H4.
