Abstract

The focus of this project was to study HECT E3 ubiquitin-protein ligases and their role as regulatory factors in the TGFbeta signaling pathway, it will focus on the regulation SMAD2 and 3 Smurf 2 is a known HECT E3 ubiquitin-protein ligase and has been shown to regulate SMAD2 and SMAD3. This project begins work to look at other members of this HECT E3 ligase family and see if they have a similar involvement as does in the TGFbeta signaling pathway Smurf2. The Approach was to construct plasmids that encoded for fusion proteins, made up of a SMAD gene and an auto fluorescing marker. Then confirmed shRNA viruses we have created to knockdown the expression of a given HECT E3. If the HECT E3 is involved in the regulation of SMAD then we will see an increase of fluoresces in knocked down cells.
