A Packed-Bed Bioreactor System for Enhancing Vero Cell Growth in a Semi-Continuous Mode of Operation

The utilization of Vero cells for vaccine production has contributed significantly to controlling viral diseases and improving the healthcare system in the US and around the world. In the United States, the adherent cell line Vero 76 has never been grown within a packed-bed bioreactor using Eppendorf’s BioFlo 320 system, however they are typically grown on microcarrier beads in a pitched-blade vessel. I compare a glass vessel to a single use vessel running in a semi-continuous mode of operation with daily glucose uptake and lactate production samples. The objective of this study was to determine if a packed-bed bioreactor can be seeded at a lower than typical density to ultimately decreases time, cost, and human risk during the seed train and scale-up process.
