Genomic Tagging of the Anaphase-Promoting Complex Activator Protein Cdc20 in S. cerevisiae

To facilitate studies of HTLV-1 protein Tax interaction with the APC/C activator protein Cdc20 in S. cerevisiae, a 9 amino acid hemagglutinin epitope tag was attached to the C terminus of the Cdc20 protein using homologous recombination of the yeast genome. A PCR product was engineered to contain homologous arms that would facilitate recombination of the tag, as well as a TRP1 marker, into the cdc20 locus. Integrants were selected on Trp- media and positive clones were identified using western blotting.
