Abstract

CopA P1B-ATPase from A. fulgidus transports Cu/Ag. The Cysteines in the CPC sequence in H6 are involved in metal coordination. However, the mechanism of metal selectivity remains unknown. To elucidate their role in in metal transport, five conserved residues were mutated by site directed mutagenesis. Two mutants from H7 (Y682A, N683A) and one from H8 (M711A) are inactivated; these residues participate in metal coordination. Mutants N675A and P688A retain partial activity and metal affinity; these residues do not participate in metal coordination.
