Abstract

The purpose of this project was to develop a strategy based on guided adaptors for targeted entry (GATEs) technology to allow the targeted transduction of cell types of interest. In this project, the GATEs system was utilized in conjunction with ALV-pseudotyped lentivirus to target gene delivery to neurons by fusing the ligand nerve growth factor (NGF) to the N-terminus of the TVA receptor (for Avian Leukosis Virus subgroup A). The primary focus of this project was to determine if the fusion of NGF to the N-terminus of TVA results in a viable TVA receptor, since there is no precedence in the literature for the activity of TVA N-terminal fusion. The results indicate that the fusion of NGF to the N-terminus of TVA result in a viable TVA receptor since viral entry did occur.
