Abstract

The focus of this project was to gain understanding on the putative copper importer PA5030 in the opportunistic bacteria Pseudomonas aeruginosa and its relationship to other proteins in the copper homeostasis network. First, recombination techniques were employed to successfully develop a double knockout mutant of the PA5030 gene and a different copper importer, PA1848 (ccoA). Second, in-silico exploration of the structure of the protein, accompanied by the development of a homolog filtering script, revealed two potential copper binding sites, M157-M173-H355 and C65-M185-M262, and 925 homologs that further suggest that PA5030 is a copper transporter.
